The Nsenkyire Stadium erupted in spontaneous celebration, with festivities expected to continue late into the night.
Samartex only needed to avoid defeat to claim the title after second-place Aduana FC lost to Bechem United on Saturday.
The dethroned champions, Medeama, also fell short after a 1-0 home defeat to Heart of Lions, paving the way for Samartex to win the Premier League grandly.
With this victory, the Timber Giants now hold a nine-point lead at the top of the table with just two matches remaining in the season.
